PLoS ONE 10(11): e0142786. doi: 10.1371/journal.pone.0142786 PMID: 26571146 Fig 4. Zonisamide rescues cell death due to oxidative stress. Primary motor neurons in DMEM/F12 with 0.5% FBS were treated with variable concentrations of zonisamide. After 1 h, cells were exposed to 100 μM hydrogen peroxide (H2O2) for 24 h. The number of viable cells was estimated by the MTS assay and was normalized to that without H2O2 (control). Mean and SE are indicated (n = 6). *p < 0.05 by one-way ANOVA followed by Tukey HSD.

Zonisamide Enhances Neurite Elongation of Primary Motor Neurons and Facilitates Peripheral Nerve Regeneration In Vitro and in a Mouse Model

No clinically applicable drug is currently available to enhance neurite elongation after nerve injury. To identify a clinically applicable drug, we screened pre-approved drugs for neurite elongation in the motor neuron-like NSC34 cells. We found that zonisamide, an anti-epileptic and anti-Parkinson's disease drug, promoted neurite elongation in cultured primary motor neurons and NSC34 cells in ...

In vitro development of a tissue-engineered model of peripheral nerve regeneration to study neurite growth.

A unique tissue-engineered model of peripheral nerve regeneration was developed in vitro to study neurite outgrowth. Mouse dorsal root ganglia neurons were seeded on a collagen sponge populated with human endothelial cells and/or human fibroblasts. Addition of nerve growth factor (NGF; 10 ng/ml) was not required for sensory neurons survival but was necessary to promote neurite outgrowth, as ass...

Sphingosine-1-Phosphate and the S1P3 Receptor Initiate Neuronal Retraction via RhoA/ROCK Associated with CRMP2 Phosphorylation

The bioactive lipid sphingosine-1-phosphate (S1P) is an important regulator in the nervous system. Here, we explored the role of S1P and its receptors in vitro and in preclinical models of peripheral nerve regeneration. Adult sensory neurons and motor neuron-like cells were exposed to S1P in an in vitro assay, and virtually all neurons responded with a rapid retraction of neurites and growth co...

Effects of Valproic Acid on Axonal Regeneration and Recovery of Motor Function after Peripheral Nerve Injury in the Rat

Background:   Valproic acid (VPA) is used to be an effective anti-epileptic drug and mood stabilizer. It has recently been demonstrated that VPA could promote neurite outgrowth, activate the extracellular signal regulated kinase pathway, and increases bcl-2 and growth cone-associated protein 43 levels in spinal cord. In the present research we demonstrate the effect of VPA on peripheral nerve r...

Localization of Epidermal-Type Fatty Acid Binding Protein (E-FABP) in Degeneration and Regeneration of Sciatic Nerve after Crush Injury in Mouse

Purpose:The regeneration of axon and myelin sheet after crush injury of peripheral nerves involves interaction of several types of cells, including Schwann cells, monocyte, macrophage and fibroblast. Among them, haematogenous macrophages invading into the peripheral nervous systein play a major role in myelin uptake during Wallerian degeneration. Materials and Methods: In this study 35 C57/BL6 ...
